The compound is registered in Russia as a pharmaceutical product, most commonly formulated as a nasal solution, and has been used in that setting since the 1990s. Outside that jurisdiction it is generally handled as a research chemical rather than an approved medicine. Regulatory status therefore differs sharply between countries, and material sold internationally may not correspond to the Russian pharmaceutical formulation. Documentation with commercial samples is typically limited to a certificate of analysis covering purity and identity, not clinical status or local legal classification.
Semax is a synthetic heptapeptide with the sequence Met-Glu-His-Phe-Pro-Gly-Pro. It corresponds to the ACTH(4-7) fragment extended at the C-terminus by a Pro-Gly-Pro tripeptide, an addition intended to slow enzymatic breakdown. The molecular formula is C37H51N9O10S and the molecular mass is approximately 814 daltons. In the literature it is often described as an ACTH(4-10) analog, although that label reflects a naming convention as much as a precise structural relationship. The compound was developed in Russia and remains most closely associated with that research tradition.
Identity and purity are commonly assessed by reversed-phase high-performance liquid chromatography, with ultraviolet detection near 214 nanometers for the peptide backbone. Mass spectrometry, either electrospray or matrix-assisted laser desorption, confirms molecular mass and detects truncation or modification products. Amino acid analysis can verify composition. Because the sequence contains no strongly absorbing aromatic residue apart from phenylalanine, detection wavelengths are chosen carefully. Purity values above 95 percent are typical for research-grade material.
Quality documentation for research-grade peptide usually includes a certificate of analysis stating purity, identity, and residual counterion content. Batch-to-batch variation in purity and salt form is a recognized issue, and comparisons across suppliers require attention to the exact counterion, for example acetate versus trifluoroacetate. Sequence verification by tandem mass spectrometry provides stronger evidence than a single mass measurement. For any study reporting biological results, the analytical method and the measured purity should be stated explicitly. Independent verification of supplier claims is considered good practice.
Semax is supplied as a lyophilized powder that appears white to off-white. It dissolves readily in water, phosphate-buffered saline, and other aqueous media, which simplifies preparation of working solutions for laboratory use. The nasal products registered in Russia are dilute aqueous solutions, typically around 0.1 percent peptide by weight. Organic solvents are rarely necessary and can complicate handling. Because the peptide is hygroscopic, weighing should be performed quickly and with minimal exposure to ambient humidity.

Clinical reports describe use in ischaemic stroke, transient ischaemic attack, optic nerve conditions, and cognitive complaints, but most of these studies are small and were conducted in a single region. Systematic reviews have generally described the evidence base as limited in size and variable in methodological quality. Randomised controlled data suitable for international regulatory assessment are scarce. As a result, major treatment guidelines outside Russia do not include the peptide, and interest in it remains largely research-driven rather than routine clinical.
Semax is a synthetic heptapeptide with the sequence Met-Glu-His-Phe-Pro-Gly-Pro. It is described in the literature as an analogue of the ACTH(4–10) fragment, a short N-terminal portion of adrenocorticotropic hormone that retains some neurotropic activity without the full hormonal effects of the parent peptide. The molecule carries a methionine residue at the N-terminus and two proline residues near the C-terminus, features that shape both its interactions with receptor systems and its chemical stability. The free peptide corresponds to the formula C37H51N9O10S and a molecular mass near 813.9 Da.
Claims about enhanced focus, memory, or mood in healthy people rest mostly on anecdotal reports and community discussion rather than on controlled data. It remains unclear whether any cognitive benefit observed in patients recovering from brain injury would extend to uninjured users. Dose-response relationships, long-term safety, and interactions with other drugs are not well characterized in the peer-reviewed literature. Questions about optimal route of administration and treatment duration likewise remain open.
Semax binds to melanocortin receptors and is thought to influence neuronal survival and plasticity rather than to act through the adrenal axis. Laboratory work has shown increased expression of brain-derived neurotrophic factor and nerve growth factor in treated tissue. Changes in c-Fos, a marker of neuronal activation, have also been reported. Because the peptide is rapidly degraded by peptidases, its effects are generally attributed to downstream signaling cascades rather than to sustained receptor occupancy.
Regulatory status differs sharply between jurisdictions. In Russia the peptide is registered as a prescription nasal preparation, while agencies such as the United States Food and Drug Administration have not approved it for any indication. Products sold elsewhere are typically labeled for laboratory research only, and such labels shift responsibility for safe handling to the purchaser. Because the same name covers pharmaceutical-grade nasal drops and bulk research powder, identity and purity documentation becomes the main practical concern when comparing sources.
Semax is a synthetic seven-amino-acid peptide whose sequence extends the ACTH(4-10) fragment with a C-terminal proline-glycine-proline tripeptide. The commonly cited sequence is Met-Glu-His-Phe-Pro-Gly-Pro, giving a molecular formula near C37H51N9O10S and a molecular weight close to 813.9 g/mol. It belongs to the broader class of synthetic ACTH fragments studied for central nervous system effects rather than for adrenal steroid stimulation. In practice the material appears as a lyophilized white powder for laboratory work or as a dilute saline solution in clinical settings.
Development is attributed to researchers at the Institute of Molecular Genetics in Moscow during the early 1980s, building on earlier Soviet work with ACTH fragments. Russian regulatory approval followed for intranasal use, and the compound has remained commercially available there for decades. Most published human data originate from Russian and, later, some Eastern European clinical reports, which are not always accessible in English translation. Outside that region the material is generally handled as a research chemical rather than a licensed medicine.
pinocytosis A form of endocytosis in which liquid and suspended solids from the extracellular environment are captured in inward invaginations of the cell membrane which then "bud off" into enclosed vesicles in the cytoplasm. The contents of these vesicles are then passed to organelles such as endosomes by fusion of the vesicular and organellar membranes. Pinocytosis is the predominant form of endocytosis occurring in most cells, such that the term is often used interchangeably with endocytosis as a whole.
The Tolkiens had four children: John Francis Reuel Tolkien (17 November 1917 – 22 January 2003), Michael Hilary Reuel Tolkien (22 October 1920 – 27 February 1984), Christopher John Reuel Tolkien (21 November 1924 – 16 January 2020) and Priscilla Mary Anne Reuel Tolkien (18 June 1929 – 28 February 2022). Tolkien was very devoted to his children and sent them illustrated letters from Father Christmas when they were young.

== Signs and symptoms == Initially, the symptoms of biliary atresia are indistinguishable from those of neonatal jaundice, a usually harmless condition commonly seen in infants. However, infants with biliary atresia develop progressive conjugated jaundice, pale white stools, dark urine, and an enlarged palpable liver. Some infants fail to thrive as there will be a degree of fat and fat-soluble vitamin malabsorption (e.g. Vitamin K). This may cause a bleeding tendency. Eventually, and usually after 2 months, cirrhosis with portal hypertension will develop. If left untreated, biliary atresia can lead to liver failure. Unlike other forms of jaundice, however, biliary-atresia-related cholestasis mostly does not result in kernicterus, a form of brain damage resulting from liver dysfunction. This is because in biliary atresia, the liver, although diseased, is still able to conjugate bilirubin, and conjugated bilirubin is unable to cross the blood–brain barrier.

== History == Dimethocaine was originally synthesized by the Hoffmann-La Roche company in 1930. It was sold under the market name larocaine. During the 1930s dimethocaine gained popularity in the US as a local anesthetic. Just like cocaine and procaine, it was used during surgery, primarily in dentistry, ophthalmology and otolaryngology. However, in the 1940s, it was removed from the market because of its psychoactive effects and risk of addiction. Nowadays dimethocaine is abused for these psychoactive effects. It is sold as a cocaine surrogate to circumvent legislation issues.
The "mirror world" would contain copies of all existing fundamental particles. On 2 May, another twin/pair-world or "bi-worlds" cosmology is shown to theoretically be able to solve the cosmological constant (Λ) problem, closely related to dark energy: two interacting worlds with a large Λ each resulting in a small shared effective Λ. Previous similar models e.g. attempt to explain the baryon asymmetry – why there was more matter than antimatter at the beginning – with a mirror anti-universe. 19 May Scientists report that RNA was found to be formed spontaneously on prebiotic basalt lava glass which is presumed to have been abundantly available on the early Earth. In normal-phase TLC, the stationary phase is polar. Silica gel is very common in normal-phase TLC. More polar compounds in a sample mixture interact more strongly with the polar stationary phase. As a result, more-polar compounds move less (resulting in smaller RF) while less-polar compounds move higher up the plate (higher RF). A more-polar mobile phase dissolves polar compounds more. As such, all compounds on the TLC plate move higher up the plate in polar solvent mixtures. "Strong" solvents move compounds higher up the plate, whereas "weak" solvents move them less. If the stationary phase is non-polar, like C18-functionalized silica plates, it is called reverse-phase TLC. In this case, non-polar compounds move less and polar compounds move more. The solvent mixture will also be much more polar than in normal-phase TLC.
The Armenian-American radiologist Mihran Krikor Kassabian (1870-1910), vice president of the American Roentgen Ray Society (ARRS), was concerned about the irritating effects of X-rays. In a publication, he mentioned his increasing problems with his hands. Although Kassabian recognized X-rays as the cause, he avoided making this reference so as not to hinder the progress of radiology. In 1902, he suffered a severe radiation burn on his hand. Six years later, the hand became necrotic and two fingers of his left hand were amputated. Kassabian kept a diary and photographed his hands as the tissue damage progressed. He died of cancer in 1910.

== Functions == Endorphins play a major role in the body's inhibitory response to pain. Research has demonstrated that meditation by trained individuals can be used to trigger endorphin release. Laughter may also stimulate endorphin production and elevate one's pain threshold. Endorphin production can be triggered by vigorous aerobic exercise. The release of β-endorphin has been postulated to contribute to the phenomenon known as "runner's high". However, experimental blockade of the mu opioid receptor has shown that endorphins are not strictly necessary for the development of exercise-induced euphoria. It has been suggested that other neurotransmitters, namely endocannabinoids, are more likely contributors. Endorphins may partially mediate exercise-induced analgesia.
== Genetics == The genetics of Bruck syndrome differs from osteogenesis imperfecta. Osteogenesis imperfecta usually involves autosomal dominant mutations to COL1A1 or COL1A2 which encode type 1 procollagen. Bruck syndrome is linked to mutations in two genes, and therefore is divided in two types. Bruck syndrome type 1 is caused by a homozygous mutation in the FKBP10 gene. Type 2 is caused by a homozygous mutation in the PLOD2 gene.
In 2013 for the population aged 40-80 years, the global prevalence of glaucoma was estimated at 3.54%, thus affecting 64.3 million worldwide. The same year, 2.97 million people in North America had open-angle glaucoma. By 2040, the prevalence of all types of glaucoma was projected to increase to 111.82 million worldwide and to 4.72 million in North America. Globally, glaucoma is the second-leading cause of blindness, while cataracts are a more common cause. In the United States, glaucoma is a leading cause of blindness for African Americans, who have higher rates of primary open-angle glaucoma, and Hispanic Americans. Bilateral vision loss can negatively affect mobility and interfere with driving. A meta-analysis published in 2009 found that people with primary open-angle glaucoma do not have increased mortality rates or increased risk of cardiovascular death. A 2024 JAMA Ophthalmology article reports that in 2022, an estimated 4.22 million people in the U.S. had glaucoma, with 1.49 million experiencing vision impairment due to the condition, according to a meta-analysis. The study found that Black adults were about twice as likely to be affected by glaucoma as White adults. Glaucoma prevalence was 1.62% among individuals aged 18 and older and 2.56% among those aged 40 and older, while vision-affecting glaucoma occurred in 0.57% and 0.91% of these age groups, respectively.
=== Acute bone fracture care === Bone fractures are treated in individuals with osteogenesis imperfecta in much the same way as they are treated in the general population; OI bone heals at the same rate as non-OI bone. A greater emphasis is placed on using lightweight materials to immobilize the fracture, as in moderate or severe types of OI, using heavy casts, such as hip spica casts, can cause fractures at the bones at the boundaries of the cast, as well as generalized osteopenia. The lightweight cast or splint is then replaced with a removable orthosis after a few weeks and once evidence of union is seen on X-ray. To prevent a nonunion or malunion, all fractures should be immobilized, even if the fracture seems trivial (microfracture), as people with OI are at greater risk of nonunion. Bone infections secondary to fractures are treated as and when they occur with the appropriate antibiotics and antiseptics, as in the general population.
A DNA triple helix is made up of three separate DNA strands, each oriented with the sugar/phosphate backbone on the outside of the helix and the bases on the inside of the helix. The bases are the part of the molecule closest to the triple helix's axis, and the backbone is the part of the molecule farthest away from the axis. The third strand occupies the major groove of relatively normal duplex DNA. The bases in triplex DNA are arranged to match up according to a Hoogsteen base pairing scheme. Similarly, RNA triple helices are formed as a result of a single stranded RNA forming hydrogen bonds with an RNA duplex; the duplex consists of Watson-Crick base pairing while the third strand binds via Hoogsteen base pairing.

It is registered as a pharmaceutical product in Russia, where it is typically supplied as a nasal solution. In most other countries it is not an approved drug and is traded as a research chemical instead. Approval status depends entirely on the jurisdiction involved.
Most published work uses intranasal administration, matching the route of the registered Russian formulation. Some animal experiments use intravenous or intraperitoneal injection. The route matters because the intact peptide is cleared rapidly from blood, so exposure differs between methods.
A purity value describes the share of the powder accounted for by the target peptide, usually by high-performance liquid chromatography. It does not confirm that the sequence is correct, which requires mass spectrometry or amino acid analysis. Purity and identity are separate checks.
Solid peptide is best kept frozen at about minus 20 degrees Celsius in a sealed container with desiccant. It should be allowed to reach room temperature before opening to prevent condensation. Repeated warming and cooling of the same vial is discouraged.
