This usually occurs by a carefully regulated series of epigenetic modifications which change the specific set of genes expressed by the cell, turning certain genes "off" and others "on". These modifications result in a cascade of phenotypic changes which can dramatically alter the cell's size, shape, metabolism, membrane properties, and rate of division, and therefore its functions, behaviors, and responsiveness to signals, permitting multicellular organisms to create a huge variety of functionally distinct cell types from a single genome. Differentiation occurs repeatedly during an organism's development from a single-celled zygote into a complex multicellular system of tissues and cell types, and continues to some extent after the organism reaches maturity in order to repair and replace damaged and dying cells. They found that 3 of the 39 substitutions significantly reduced the fitness of the ancestral background. Compensatory mutations are new mutations that arise and have a positive or neutral impact on a populations fitness. Previous research has shown that populations have can compensate detrimental mutations. Burch and Chao tested Fisher's geometric model of adaptive evolution by testing whether bacteriophage φ6 evolves by small steps. Their results showed that bacteriophage φ6 fitness declined rapidly and recovered in small steps . Viral nucleoproteins have been shown to avoid cytotoxic T lymphocytes (CTLs) through arginine-to glycine substitutions. This substitution mutations impacts the fitness of viral nucleoproteins, however compensatory co-mutations impede fitness declines and aid the virus to avoid recognition from CTLs. Mutations can have three different effects; mutations can have deleterious effects, some increase fitness through compensatory mutations, and lastly mutations can be counterbalancing resulting in compensatory neutral mutations.

On 1 June 2020, the United States Pharmacopeia (USP) decided to cancel the proposal to include recombinant technology for endotoxin testing in chapter 85, Bacterial Endotoxins, and start the development of a separate chapter that expands on the use, validation, and comparability of endotoxin tests based on recombinantly derived reagents. A separate guidance-only chapter 1085.1 was proposed by the USP, though comments and feedback published on 11 December 2020 show that pharmaceutical companies and the FDA did not support this chapter and requested compendial status. In response, USP subsequently developed General Chapter <86>, Bacterial Endotoxins Test Using Recombinant Reagents, which provides compendial requirements for endotoxin testing using non-animal-derived reagents such as recombinant factor C (rFC). Chapter <86> became official in USP–NF in May 2025, establishing rFC as a pharmacopeially recognized alternative to the Limulus amebocyte lysate test.
== Treatment == The treatment is based on addressing obesity, thus reducing insulin resistance and its undesired effects. Insulin resistance can be treated with metformin and may have a positive impact on reproductive function. Pharmacological treatment by suppression of gonadotropin with estrogen-progesterone oral contraceptives can reduce the hyperandrogenism by decreasing LH (luteinizing hormone) levels. Even their sex hormone binding to globulin increase is also responsible for decreasing body's bio-availability of testosterone. Progestin treatment with desogestrel and norgestimate appears to have fewer androgenic side effects and may be safer to use in persons with abnormal lipid levels or hirsutism. Other proposed treatments include antiandrogenic medications, spironolactone (in combination with oral contraceptives to prevent menstrual cycle irregularities), flutamide, and the 5α-reductase inhibitor finasteride.

Within complex communities such as Bush Hill, petroleum seems less important than previously thought (MacDonald, 1998b). Through taphonomic studies (death assemblages of shells) and interpretation of seep assemblage composition from cores, Powell et al. (1998) reported that, overall, seep communities were persistent over periods of 500–1,000 years and probably throughout the entire Pleistocene. Some sites retained optimal habitat over geological time scales. Powell reported evidence of mussel and clam communities persisting in the same sites for 500–4,000 years. Powell also found that both the composition of species and trophic tiering of hydrocarbon seep communities tend to be fairly constant across time, with temporal variations only in numerical abundance. He found few cases in which the community type changed (from mussel to clam communities, for example) or had disappeared completely. Faunal succession was not observed. Surprisingly, when recovery occurred after a past destructive event, the same chemosynthetic species reoccupied a site. There was little evidence of catastrophic burial events, but two instances were found in mussel communities in Green Canyon Block 234. The most notable observation reported by Powell (1995) was the uniqueness of each chemosynthetic community site.

For many black Detroiters, the dismissal of FHA redlining did not end the economic exploitation and residential segregation of black families at the hands of wealth-hungry bankers and lenders. Colorblind universalism within Detroit's housing market remained a far-fetched aspiration of Black families in their pursuit of homeownership as FHA-HUD policy failed to dismantle the segregative impulses of the real estate industry. Furthermore, the threat of racial integration in white communities facilitated the rise of neighborhood associations, which were coalitions of independent white homeowners that fervently protected homeownership rights through advocacy for residential segregation.
== Causes == Cholecystitis occurs when the gallbladder becomes inflamed. Gallstones are the most common cause of gallbladder inflammation but it can also occur due to blockage from a tumor or scarring of the bile duct. The greatest risk factor for cholecystitis is gallstones. Risk factors for gallstones include female sex, increasing age, pregnancy, oral contraceptives, obesity, diabetes mellitus, ethnicity (Native North American), rapid weight loss.
(1) Most often, 5'-DMT group is removed at the end of the oligonucleotide chain assembly. The oligonucleotides are then released from the solid phase and deprotected (base and phosphate) by treatment with aqueous ammonium hydroxide, aqueous methylamine, their mixtures, gaseous ammonia or methylamine or, less commonly, solutions of other primary amines or alkalies at ambient or elevated temperature. This removes all remaining protection groups from 2'-deoxyoligonucleotides, resulting in a reaction mixture containing the desired product. If the oligonucleotide contains any 2'-O-protected ribonucleotide residues, the deprotection protocol includes the second step where the 2'-O-protecting silyl groups are removed by treatment with fluoride ion by various methods. The fully deprotected product is used as is, or the desired oligonucleotide can be purified by a number of methods. Most commonly, the crude product is desalted using ethanol precipitation, size exclusion chromatography, or reverse-phase HPLC. To eliminate unwanted truncation products, the oligonucleotides can be purified via polyacrylamide gel electrophoresis or anion-exchange HPLC followed by desalting. (2) The second approach is only used when the intended method of purification is reverse-phase HPLC. In this case, the 5'-terminal DMT group that serves as a hydrophobic handle for purification is kept on at the end of the synthesis. The oligonucleotide is deprotected under basic conditions as described above and, upon evaporation, is purified by reverse-phase HPLC.
