An enzyme inhibitor is characterised by its dissociation constant Ki, the concentration at which the inhibitor half occupies the enzyme. In non-competitive inhibition the inhibitor can also bind to the enzyme-substrate complex, and the presence of bound substrate can change the affinity of the inhibitor for the enzyme, resulting in a second dissociation constant Ki'. Hence Ki and Ki' are the dissociation constants of the inhibitor for the enzyme and to the enzyme-substrate complex, respectively. === Genomes === Genomic DNA is tightly and orderly packed in the process called DNA condensation, to fit the small available volumes of the cell. In eukaryotes, DNA is located in the cell nucleus, with small amounts in mitochondria and chloroplasts. In prokaryotes, the DNA is held within an irregularly shaped body in the cytoplasm called the nucleoid. The functional genetic information in a genome is located in genes, regulatory sequences, origins of replication, centromeres, telomeres, and segments required for the three-dimensional structure of chromatin. In many complex eukaryotes, only a small fraction of the total sequence is devoted to the various functional elements. == Preparedness == A number of countries conduct exercise to increase preparedness and explore the strategy, tactics and operations involved in conducting and defending against cyber attacks against hostile states, this is typically done in the form of war games. The Cooperative Cyber Defence Centre of Excellence (CCDCE), part of the North Atlantic Treaty Organization (NATO), have conducted a yearly war game called Locked Shields since 2010 designed to test readiness and improve skills, strategy tactics and operational decision making of participating national organizations. Locked Shields 2019 saw 1200 participants from 30 countries compete in a red team vs. blue team exercise. The war game involved a fictional country, Berylia, which was "experiencing a deteriorating security situation, where a number of hostile events coincide with coordinated cyber attacks against a major civilian internet service provider and maritime surveillance system. The attacks caused severe disruptions in the power generation and distribution, 4G communication systems, maritime surveillance, water purification plant and other critical infrastructure components". CCDCE describe the aim of the exercise was to "maintain the operation of various systems under intense pressure, the strategic part addresses the capability to understand the impact of decisions made at the strategic and policy level." Ultimately, France was the winner of Locked Shields 2019.
== Construction == The gene on a DNA sequence of interest can either be cloned from an existing sequence or developed synthetically. To clone a naturally occurring sequence in an organism, the organism's DNA is first cut with restriction enzymes, which recognize DNA sequences and cut them, around the target gene. The gene can then be amplified using polymerase chain reaction (PCR). Typically, this process includes using short sequences known as primers to initially hybridize to the target sequence; in addition, point mutations can be introduced in the primer sequences and then copied in each cycle in order to modify the target sequence. It is also possible to synthesize a target DNA strand for a DNA construct. Short strands of DNA known as oligonucleotides can be developed using column-based synthesis, in which bases are added one at a time to a strand of DNA attached to a solid phase. Each base has a protecting group to prevent linkage that is not removed until the next base is ready to be added, ensuring that they are linked in the correct sequence. Oligonucleotides can also be synthesized on a microarray, which allows for tens of thousands of sequences to be synthesized at once, in order to reduce cost. To synthesize a larger gene, oligonucleotides are developed with overlapping sequences on the ends and then joined together. The most common method is called polymerase cycling assembly (PCA): fragments hybridize at the overlapping regions and are extended, and larger fragments are created in each cycle.

If Hydra are alarmed or attacked, the tentacles can be retracted to small buds, and the body column itself can be retracted to a small gelatinous sphere. Hydra generally react in the same way regardless of the direction of the stimulus, and this may be due to the simplicity of the nerve nets. Hydra are generally sedentary or sessile, but do occasionally move quite readily, especially when hunting. They have two distinct methods for moving: looping and somersaulting. They do this by bending over and attaching themselves to the substrate with the mouth and tentacles and then relocate the foot, which provides the usual attachment; this process is called looping. In somersaulting, the body then bends over and makes a new place of attachment with the foot. By this process of looping or somersaulting, a Hydra can move several inches (c. 100 mm) in a day. Hydra may also move by amoeboid motion of their bases or by detaching from the substrate and floating away in the current. A dark-habituated Hydra that is exposed to light will respond by elongating its body towards it, bending its hypostome-tentacle junction, and eventually somersaulting towards the light source.
=== Hyperthermia therapy === Hyperthermia therapy is heat treatment for cancer that can be a powerful tool when used in combination with chemotherapy (thermochemotherapy) or radiation for the control of a variety of cancers. The heat can be applied locally to the tumor site, which will dilate blood vessels to the tumor, allowing more chemotherapeutic medication to enter the tumor. Additionally, the tumor cell membrane will become more porous, further allowing more of the chemotherapeutic medicine to enter the tumor cell. Hyperthermia has also been shown to help prevent or reverse "chemo-resistance." Chemotherapy resistance sometimes develops over time as the tumors adapt and can overcome the toxicity of the chemo medication. "Overcoming chemoresistance has been extensively studied within the past, especially using CDDP-resistant cells. In regard to the potential benefit that drug-resistant cells can be recruited for effective therapy by combining chemotherapy with hyperthermia, it was important to show that chemoresistance against several anticancer drugs (e.g. mitomycin C, anthracyclines, BCNU, melphalan) including CDDP could be reversed at least partially by the addition of heat.

== Description == Typical Catostylus with chunky appendages and tentacle to go with each. This jellyfish has a sting that causes light pain and a skin rash, but generally poses no serious threat. It is up to 65 cm (26 in) in bell diameter, but a more typical size is 25 cm (10 in) in diameter and 2.5 kg (5.5 lb) in weight. Its colour is variable and can be blue-white, cream, brown, or off-white. The exumbrellar grooves are reddish or purplish brown. C. tagi has gonads along the edge of its stomach in an X shape. It has the octant formation typical of Catostylus jellies, the height of the octants are also variable.
