=== Neuropathy === Neuropathy has been a problem in some clinical trials with DCA causing them to be effectively halted, but a 2008 BJC review found that it has not occurred in other DCA trials. The mechanism of DCA induced neuropathy is not well understood. On the one hand in vitro work with nerves has suggested a mechanism for the neuropathic effect of DCA; with DCA showing a dose and exposure dependent demyelination of nerves (stripping of the nerve 'sheath'), which demyelination was partially reversible over time, following washout of DCA. On the other hand, the 2008 review in BJC states "This neurotoxicity resembled the pattern of length-dependent, axonal, sensorimotor polyneuropathy without demyelination." with regard to the 2006 study by Kaufman et al.
Peyote cacti were studied scientifically in the 1800s, culminating in the isolation of mescaline as the active psychedelic constituent in cacti, legal recognition of religious use, and ongoing exploration of the therapeutic potential of mescaline and other analogues. Mescaline was the first psychedelic compound to be characterized by the Western world and has been referred to as the "first psychedelic". Mescaline and the cacti that produce it are largely illegal worldwide, though exceptions exist for religious, scientific, or ornamental use, and the compound has influenced many notable cultural figures, for instance Aldous Huxley and Alexander Shulgin among others. ==== Addiction ==== Whether caffeine can result in an addictive disorder depends on how addiction is defined. Compulsive caffeine consumption under any circumstances has not been observed, and caffeine is therefore not generally considered addictive. Some diagnostic sources, such as the ICD-9 and ICD-10, include a classification of caffeine addiction under a broader diagnostic model. Caffeine does not appear to be a reinforcing stimulus, and some degree of aversion may actually occur, with people preferring placebo over caffeine in a study on drug abuse liability published in an NIDA research monograph. Some state that research does not provide support for an underlying biochemical mechanism for caffeine addiction. Other research states it can affect the reward system. "Caffeine addiction" was added to the ICD-9 and ICD-10. However, its addition was contested with claims that this diagnostic model of caffeine addiction is not supported by evidence. The American Psychiatric Association's DSM-5 does not include the diagnosis of a caffeine addiction but proposes criteria for the disorder for more study. === Songwriting process === In charge of the basic structure of the songs, Chanut or Thépegnier would then present them in rehearsal, where the other band members would develop them through a "more organic" process. Toufouti usually writes the lyrics after receiving the instrumentals. Chanut said in 2015 that he "validates his lyrics so that they totally align with the band's concept". Toufouti writes his lyrics in English for convenience. According to him, it would be more difficult in French, because of the risk of falling into excess and clichés. Furthermore, according to Chanut, the inherent fluidity of English is better suited to metal music, whereas certain French consonants are more abrupt. He added that most of the bands they listen to sing in English. Conversely, Hangman's Chair enjoys using French album or song titles for their sonority. Chanut and Thépegnier had to adapt to periods of lockdown while the rehearsal space remained closed for more than a year. They were both able to exchange ideas, as Thépegnier had his home studio under construction and Chanut had his own audio workstation. They were also able to rehearse remotely without audio latency thanks to the beta version of the NuCorder software, designed by Caste. The drum patterns that would later appear on A Loner were influenced by Thépegnier's use of an electronic drum kit during these remote songwriting sessions. == Mode of action == The exact activation mechanism of Ppk1/Ppk26 ion channels in the presence of Do6a is unknown; however, it likely compares to the mode of action of endogenous peptide Vulnusin due to their similarity in structure. Vulnusin binds to Ppk/Bba channels upon mechanical injury, yet administration of this peptide independently still results in channel activation and nociceptive rolling in Drosophila larvae. This could mean that a wound in larvae tissue allows Vulnusin, normally located above the epidermal layer, to reach its receptors below epidermal cells. Do6a, delivered to Ppk/Bba receptors through a mechanical puncture of the tissue from the velvet ant sting, could elicit its response through a similar mechanism. Since Ppk1/Ppk26 ion channels are homologous to Acid-sensing ion channels (ASICs) in vertebrates, they are likely to be permeable to cations, however, the exact ion type that passes through these channels upon activation has not been determined.
