The first and most important step in sample preparation for mass spectrometry is determining what phase the sample needs to be in. Different ionization methods require different sample phases. == Clinical significance == Mutations of the CELA1 gene were suspected to be associated with diffuse nonepidermolytic palmoplantar keratoderma (diffuse NEPPK). However the suspected sequence variant was fully functional and did not strongly associate with the disease. More recently, a specific mutation in the KRT6C gene has been linked to some cases of diffuse NEPPK. A possible polymorphism of the CELA1 gene coding this protein was found. On a secondary structure level, this polymorphism manifests itself in an excision of a short sequence of CELA1. The disappeared sequence carries the key amino acid residues Val-227 and Thr-239, which contribute to the substrate specificity of elastase I (highlighted in Figure 3), as well as five of the eight amino acids involved in the primary contact of the elafin(inhibitor)/elastase complex formation. These observations imply that the sequence variant might modify the substrate specificity of the enzyme and abolish the inhibitor binding capability. Though there were no obvious pathogenic epidermal abnormalities associated with the truncated ELA1 variant, it is possible that carriers of the polymorphism may be at greater risk of developing the common skin diseases such as psoriasis and eczema (genetic and histologic studies will be required to investigate the role of ELA1 in these common epidermal disorders.).
== General technique for protein purification == A slurry of resin, such as DEAE-Sephadex is poured into the column. The matrix that is used is insoluble with charged groups that are covalently attached. These charged groups are referred to as exchangers like cation and anion exchangers. After it settles, the column is pre-equilibrated in buffer before the protein mixture is applied. DEAE-Sephadex is a positively charged slurry that will have electrostatic interactions with the negatively charged atoms, making them elute later than the positively charged molecules in the interested sample. This is a separation technique used widely to discover specific proteins, or enzymes in the body. Unbound proteins are collected in the flow-through and/or in subsequent buffer washes. Proteins that bind to the positively charged resin are retained and can be eluted in one of two ways. First, the salt concentration in the elution buffer is gradually increased. The negative ions in the salt solution (e.g. Cl−) compete with protein in binding to the resin. Second, the pH of the solution can be gradually decreased which results in a more positive charge on the protein, releasing it from the resin. Both of these techniques can displace the negatively charged protein which is then eluted into test tubes fractions with the buffer. The separation of proteins will depend on the differences in total charge. Composition of ionizable side chain groups will determine the total charge of the protein at a particular pH.

=== Argentina === In 2011, Argentina introduced a catalogue of drugs covered by its national drug traceability scheme, listing more than 3,000 drugs that require the placing of unique serial numbers and tamper-evident features on the secondary packaging. === Ventricular assist device === Collaboration between NASA, Dr. Michael DeBakey, Dr. George Noon, and MicroMed Technology Inc. resulted in a heart pump for patients awaiting heart transplants. The MicroMed DeBakey ventricular assist device (VAD) functions as a "bridge to heart transplant" by pumping blood until a donor heart is available. The pump is approximately one-tenth the size of other currently marketed pulsatile VADs. Because of the pump's small size, fewer patients developed device-related infections. It can operate up to 8 hours on batteries, giving patients the mobility to do normal, everyday activities.
